The study explains why certain patients develop severe infections after chemotherapy and points to ways of averting this side-effect

Cancer chemotherapy can be a life-saver, but it is fraught with severe side effects, among them an increased risk of infection. Until now, the major criterion for assessing this risk has been the blood cell count: if the number of white blood cells falls below a critical threshold, the risk of infection is thought to be high. A new model built by Weizmann Institute mathematicians in collaboration with physicians from the Meir Medical Center in Kfar Saba and from the Hoffmann-La Roche research center in Basel, Switzerland, suggests that for proper risk assessment, it is essential to evaluate not only the quantity of these blood cells, but also their quality, which varies from one person to another.

This research may represent an important step in the emerging field of personalized medicine, leading to a more individualized approach to chemotherapy. In particular, better precautions might need to be taken to prevent infection in high-risk patients whereas those at a low risk could be spared unnecessary preventive treatments.

The study, recently published in the Journal of Clinical Investigation, brought together the expertise of researchers from such diverse disciplines as applied mathematics, electrical engineering, oncology, immunology and pediatrics.

The new model reveals how the immune system functions under conditions of neutropenia dangerously low level of white blood cells, mainly neutrophils. In this condition, which often emerges after chemotherapy or bone marrow transplant, severe infections can develop if the immune system fails to perform the crucial function of devouring and destroying bacteria. "Our mathematical model has revealed previously unknown mechanisms responsible for the variability in the vulnerability of neutropenia patients to infections," says research leader Prof. Vered Rom-Kedar of the Weizmann Institute's Computer Science and Applied Mathematics Department.

The model suggests that in neutropenia, the tug of war between the blood cells and the bacteria cannot be explained away by the simple bacteria-to-cell ratio, nor by the threshold that the blood cell count must exceed. Rather, when neutrophil counts are low, the patient's immune system enters a fragile equilibrium described as "bistability" in mathematical terms which can easily be disrupted, with dramatic consequences, by even minute changes in bacterial concentration or neutrophil numbers. Other factors that can radically affect this equilibrium include the effectiveness of the neutrophil functioning and the permeability of tissues to bacteria, which can increase due to cancer therapy.

Thus according to the model, in healthy people, the fact that the effectiveness of neutrophils varies from one person to another usually has no significant consequences. In contrast, in patients with neutropenia, this individual variability can make a difference between life and death. This conclusion is drawn from the study based upon the blood analysis of four healthy volunteers. To use the model in the clinic, such analysis should be applied to large populations.

The model has already offered a plausible explanation for a number of medical mysteries. It helps explain, for example, why after chemotherapy, some cancer patients contract life-threatening infections even when in isolation under sterile conditions: If the neutrophils of these patients are "weak," even the smallest numbers of bacteria, for example, those present in the gut, can tilt the fragile immune balance in favor of the bacteria.

The study also explains why certain patients, following chemotherapy or a bone marrow transplant, may develop acute infections even if their neutrophil levels have returned to relatively normal levels. The chemotherapy lowers both neutrophil levels and function, making the tissues of these patients more penetrable to bacteria. The model suggests that as a result, in some patients the bacterial concentrations might increase so quickly that by the time the neutrophil counts rise back to "normal," the rapidly multiplying bacteria have already gained a head start, so that the neutrophil recovery is insufficient for overcoming the infection. This scenario may eventually also shed light on the rare cases in which acute bacterial infections develop in individuals with normal immunological function. The model suggests that in such cases, a high growth rate of unusually virulent bacteria could overcome the appropriate quantitative and qualitative neutrophil response.

Certain puzzling medical cases could be clarified thanks to the model. For example, a newborn baby treated at Meir Medical Center recovered from neutropenia even though his absolute neutrophil count (ANC) had fallen as low as 200 neutrophils per microliter of blood, whereas an adult whose ANC stood at 380 after chemotherapy died of infection. The model has suggested how such clinical parameters as the poor quality of the neutrophils might have lead to the death of the adult. In addition, the model could help doctors to understand the mechanism behind the development of severe recurrent infections in some patients. Of one thousand patients referred to Meir Medical Center because of such infections, diagnosis could be established in only one-third of the cases. Weizmann Institute's mathematical modeling suggests that at least some of the unresolved cases may have resulted from a combination of several mild defects, including variation in the function of neutrophils and other immune cells.

The study was performed by researchers with an unusual combination of backgrounds. Lead author, Weizmann Institute's mathematician Prof. Vered Rom-Kedar, specializes in the investigation of dynamic systems. The first author, Dr. Roy Malka, an electrical engineer who has become an applied mathematician, conducted this research as part of his Ph.D. studies at Weizmann; he is now conducting postdoctoral research on related subjects at Harvard Medical School. The idea for the project was first proposed by Dr. Eliezer Shochat, a senior oncologist who also has a Ph.D. in applied mathematics from Weizmann and now works in a research group at Hoffman-La Roche in Basel, Switzerland. The study was performed in collaboration with the Meir Medical Center team: Prof. Baruch Wolach, M.D., Head of the Laboratory for Leukocyte Function and of the Chair of Pediatric Immunology at Tel Aviv University's Sackler Faculty of Medicine, and laboratory manager Ronit Gavrieli, M.Sc., who performed the experiments.

Says Prof. Wolach: "Our study suggests that to achieve optimal results in applying chemotherapy, and/or in patients with innate neutrophil dysfunction, it is of value to assess the patient's neutrophils periodically, as well as the bacterial concentration. Such assessments will help reduce the morbidity and the mortality, as well as the cost, associated with unnecessary hospitalizations and the administration of expensive medications. Moreover, by cutting down on the use of antibiotics, these assessments can help in preventing the rise in antibiotic resistance."

NVIDIA announces its lowest priced Kepler cards the $229 GeForce 660 and $109 GeForce 650

NVIDIA's had some trouble shaving its Kepler GPUs down to an entry-level price point, but it looks to have put the problem behind it with the new GeForce 660 and 650 graphics cards. The company's ambition was to coax impoverished gamers clinging to DirectX9 (and to a lesser extent, 10) into switching up to this wallet-friendly pair of low-end units.

The 660 has been designed to be the "weapon of choice" for budget gamers. It'll play most games at reasonably high settings, thanks to its 2GB of RAM, 960 CUDA Cores and GPU Boost, which automatically overclocks the silicon according to the demands of your software. While we'll wait for real-world benchmarks, the company expects four-times the performance of the GeForce 9800GT, claiming games like Borderlands 2 and Guild Wars 2, in a resolution of 1,920 x 1,080 will play at frame rates of 51fps and 41fps with full 3D, respectively

The 650 is the company's self-proclaimed "gateway" into gaming, being the lowest-priced Kepler it's planning to produce. Unlike the other cards in the range, it lacks GPU Boost, but the company left six-pin power on the card, giving card makers 64W to push the "good overclocker" 1GHz units all the way to 1.2GHz. It's got 1GB of DDR5 RAM, which will apparently handle even the newest games at mid-range levels of detail with its 384 CUDA Cores. The pair are available from today, with companies like Maingear already announcing discounted desktops for them to nestle inside.

(AP) ? Philip Rivers threw a 6-yard touchdown pass to Malcom Floyd and the San Diego Chargers took a 16-6 lead over the mistake-prone Oakland Raiders through three quarters Monday night in the season opener for both teams.

The Chargers took advantage of three Raiders penalties that led to first downs on a 13-play, 90-yard drive late in the second quarter. Rivers completed a 17-yard pass to Antonio Gates on third-and-5 to move to Oakland's 7-yard line before finding Floyd on a short toss on another third down.

Gates went to the locker room briefly with a rib injury in the third quarter but returned for San Diego's next drive.

Raiders long snapper Jon Condo left with a head injury in the first half and did not return. Normally an overlooked position, the Raiders felt Condo's absence almost immediately.

Travis Goethel took over at long snapper and rolled a snap in the third quarter to punter Shane Lechler, who was gang tackled at Oakland's 39. Nate Kaeding kicked a 28-yard field goal to extend San Diego's lead to 13-6.

The Raiders offense stalled again and Goethel's next snap stayed straight, although Lechler appeared to stand closer to the line of scrimmage. Dante Rosario ran through the middle to block the punt, giving San Diego possession at Oakland's 8.

It was the first time the Raiders had a punt blocked since Oct. 8, 2006, at San Francisco. Condo had never had a punt blocked while at long snapper. The Chargers had not blocked a punt since Oct. 25, 2009, at Kansas City.

The only downside for the Chargers is they again failed to get in the end zone. Kaeding kicked a 19-yard field to put San Diego ahead 16-6 late in the third quarter.

Oakland relied on a mostly one-man show on offense.

Darren McFadden, out with a right foot injury most of last season, already caught a career-high 10 passes for 76 yards to go with 76 yards rushing for the Raiders, who settled for field goals of 51 and 20 yards by Sebastian Janikowski.

Familiar mistakes flared up again for the Raiders.

Nose tackle Tommy Kelly handed San Diego two first downs on the touchdown drive when he jumped offside on two short third downs. Tyvon Branch gave the Chargers another first down on a holding penalty.

The Raiders set an NFL record with 163 penalties last season, giving opponents 58 first downs by penalties.

Rivers avoided a sack and bought time in the pocket to find Floyd in the front of the end zone. Floyd dunked the ball through the uprights and waved his hands at the Black Hole side of the Oakland Coliseum after giving San Diego a 10-3 lead.

The Raiders nearly rallied for a tying touchdown before halftime.

On third-and-15 from San Diego's 16, McFadden took a short pass from Rivers and burst up the middle, splitting two defenders and diving a yard short of a first down with 7 seconds left. The replacement referees initially ruled McFadden had the first down until the replay booth overturned the call and set up a fourth-and-short.

The 14-play, 77-drive ended with Janikowski's 20-yard field goal.

Robert Meachem made a diving, 46-yard catch from Rivers at Oakland's 20 in the first quarter. After Rivers sailed passes to Floyd and Gates in the end zone, the Chargers settled for tying 23-yard field goal by Kaeding.

Oakland had plenty of production from the offense but no points.

Darrius Heyward-Bey took a short pass from Carson Palmer for 13 yards and McFadden caught a 9-yard pass before the drive stalled. Janikowski stayed steady despite a high snap and kicking from the infield dirt at the Coliseum ? shared by baseball's Oakland Athletics ? to boot a wobbly line drive through the uprights.

On Oakland's opening possession, Rod Streater caught Palmer's pass over the middle for what would have been a first down on third-and-4. As Streater spun around, Melvin Ingram came from behind to knock the ball lose and Antwan Barnes recovered at San Diego's 29.

But the Chargers couldn't capitalize on the turnover. Ronnie Brown caught a 4-yard pass, was stuffed for no gain and Matt Shaughnessy and David Tollefson sacked Rivers to force a punt.

The Raiders took over and Palmer led the seven-play, 33-yard drive that ended with Janikowski's field goal from the infield dirt. With the Miami Marlins getting a new ballpark this year, Oakland is the only stadium still shared by NFL and Major League Baseball teams.

The Raiders spiced up the home opener with a performance by rapper Ice Cube, who overcame a glitch on his microphone before getting a sold-out Coliseum crowd on its feet, singing lyrics and waving in unison. Gene Simmons from "Kiss" sang the national anthem, and former coach Tom Flores ignited the eternal flame in memory of owner Al Davis.

The season started the way last season ended for these two California franchises.

The Raiders would have won the AFC West had they beaten San Diego at home in the season finale. Instead, they lost 38-26 at home and missed the postseason for the ninth straight season.

Mayor of New Jersey's capital city arrested on corruption charge

(Reuters) - The embattled mayor of Trenton, New Jersey's state capital, was arrested at his home early on Monday and charged with public corruption, the FBI said.

The charge against Trenton Mayor Tony Mack involves conspiracy to obstruct commerce by extortion, said Barbara Woodruff, FBI spokeswoman in Newark, New Jersey.

The mayor has been the target of an ongoing federal investigation, and in July FBI agents staged an early morning raid on his home and searched offices in City Hall as well.

Mack has been accused by critics of nepotism and mismanagement since taking office in the crime-plagued, economically depressed city of 85,000 in 2010.

He was arrested at his home at about 6:30 a.m. EDT, the FBI said.

Also arrested on Monday was a longtime associate of Mack, Joseph Giorgianni, also for public corruption, the FBI said. Giorgianni also faces drug charges, it said.

In July, after the FBI raid on his home, Mack said he had done nothing wrong.

"We have not violated the public trust in any way," he said at the time.

An effort to recall Mack, a Democrat, failed last year when organizers did not get enough support to force a special election.

Mack was slated to appear before a federal magistrate in Trenton later on Monday, and U.S. Attorney Paul Fishman and Michael Ward, director of the FBI in New Jersey, planned to hold a news conference in Trenton later on Monday as well, the FBI said.

(NOTE:? many people refer to the sections of the act as beginning with section 600.? Thus ?Section 615? would be found at 20 U.S.C. Section 1415, etc.)? The regulations promulgated by the United States Department of Education to implement the IDEA are found at 34 C.F.R. Part 300.? Many state have adopted their own special education regulations. ?

Court decisions that interpret the IDEA issued by the courts of your state, by the United States Supreme Court, and by the federal Circuit Court of Appeals and the federal District Courts that cover your state or District are binding.? Other court opinions and hearing officer decisions issued under the Act may be cited and used if you find their reasoning to be persuasive, but they are not binding precedent.? Similarly, opinions issued by the federal Department of Education interpreting the Act provide helpful guidance, but they are also not binding precedent.

Although the IDEA and the federal regulations, and corresponding state regulations and policies, and the relevant decisions interpreting them are by far the most important sources of special education law, other statutes do sometimes become involved.? The Rehabilitation Act of 1973, 29 U.S.C.? Section 794, et. seq., commonly referred to as ?Section 504,? prohibits discrimination on the basis of disability in certain federally funded programs, including education.? The federal regulations that implement the statute are found at 34 C.F.R. Part 104.Another statute that will impact upon special education law is the No Child Left Behind Act, or ESEA, 20 U.S.C. Section 6301, et. seq., hereafter sometimes referred to as ?NCLB.?? The regulations implementing NCLB are located at 34 C.F.R. The weekend after Labor Day is typically the slowest of the year, but this weekend's grosses were down 20 percent over last year when "Contagion" opened in first place with $22.4 million. Total box office revenues are estimated at $67 million, which would make this the worst weekend at the box office since the weekend after 9/11 when revenues were $59.7 million.

"There just wasn't a strong opener," said Paul Dergarabedian, box-office analyst for Hollywood.com. "We came off a summer that ended with a whimper. There wasn't any momentum. It just comes down to the movies and the marketplace. There's wasn't some extraneous force keeping people out of the theaters. This crop of movies just didn't have that solid draw."

The bootlegging tale "Lawless" starring Shia LaBeouf, Tom Hardy and Jessica Chastain kept a lock on the No. 2 position in its second weekend with $6 million, bringing its total haul to $23.5 million, while "The Words" featuring Bradley Cooper as an aspiring writer and Zoe Saldana as his girlfriend debuted in third place with $5 million.

A pair of action sequels rounded out the top five films. The ensemble flick "The Expendables 2" earned $4.7 million in its fourth weekend, while "The Bourne Legacy" captured $4 million in its fifth outing at the box office. "Expendables 2" also won $14 million internationally in 23 territories, and "Bourne Legacy" nabbed $13 million in 49 territories.

"2016: Obama's America," a conservative film exploring the roots of President Barack Obama's political views, raised its total to $26 million in its ninth week of release, earning another $3.3 million this weekend. That now makes "2016" the second highest grossing political documentary of all time behind director Michael Moore's "Fahrenheit 9/11," which holds the top spot with $119.1 million.

"The Cold Light of Day," the weekend's only other new major release, wasn't able to see the light of the top 10. The drama starring Henry Cavill and Bruce Willis earned just $1.8 million in the No. 13 spot.

Below "The Cold Light of Day," an IMAX re-release of 1981's "Indiana Jones and the Raiders of the Lost Ark" collected $1.7 million in the No. 14 position. The film starring Harrison Ford as the iconic archaeologist is playing a limited engagement in 267 theaters.

Estimated ticket sales for Friday through Sunday at U.S. and Canadian theaters, according to Hollywood.com. Where available, latest international numbers are also included. Final domestic figures will be released Monday.

Mitt Romney says he won?t cut taxes on America's wealthy

Mitt Romney rejected claims by President Barack Obama that he would sign off on more tax breaks for the wealthy if elected president, but again declined to offer specifics on how exactly he would accomplish his goal of lowering taxes for other Americans while also balancing the budget.

In an interview with NBC's "Meet the Press," Romney said he would offset his proposed 20 percent tax cut for all Americans by eliminating loopholes and deductions for high-income earners. He argued that his plan would in effect lower taxes for middle class Americans while keeping tax rates the same for wealthy Americans.

"People at the high end, high income taxpayers, are going to have fewer deductions and exemptions. Those numbers are going to come down. Otherwise they'd get a tax break," Romney told NBC. "And I want to make sure people understand, despite what the Democrats said at their convention, I am not reducing taxes on high income taxpayers."

Romney argued that limiting deductions and exceptions would keep government revenues up and "encourage more hiring" and "encourage growth" in the economy. He said he wants to "makes sure we don't put any bigger burden on middle income people."

But asked about "the specifics of how you get into this math," Romney declined to offer any additional details, suggesting the "principals" of his plan should be enough for Americans to judge his tax proposals.

"The specifics are these which is those principles I described are the heart of my policy," Romney said. "And I've indicated as well that contrary to what the Democrats are saying I'm not going to increase the tax burden on middle-income families. It would absolutely be wrong to do that."

The Republican presidential nominee criticized GOP lawmakers in Congress?a group that includes his running mate, Rep. Paul Ryan-- for backing automatic budget cuts during debt negotiations last year. Under the agreement signed by the White House and the Republican Congress, the automatic cuts, known in Washington as "sequestration," will automatically remove $1.2 trillion from the budget unless lawmakers can find other cuts to replace them. At least half the cuts are coming in defense spending, which Romney called an "extraordinary miscalculation" on Obama's part and a "big mistake" for Republicans to agree to it."

"I thought it was a mistake on the part of the White House to propose it. I think it was a mistake for Republicans to go along with it," Romney said.

Asked if he would be willing to "cut a deal" in order to keep the country away from a "fiscal cliff," Romney indicated it's "critical to get the country back on track" but did not say how far he would go in working with Democrats.

"There's nothing wrong with the term compromise, but there is something very wrong with the term abandoning one's principles," Romney said. "And I'm going to stand by my principles. And those are I am not going to raise taxes on the American people. Our problem in our country is not that we're not paying enough taxes. It's that we're spending too much money and the economy is not growing as it could and should."

On Obama's health care law, Romney reiterated that he would repeal the law, if elected, but would replace it with his own measure that would include some of the elements from Obama's law, including coverage for those with "pre-existing conditions." He also said he also would keep rules allow young people to stay on their parents health care plans.

"I say we're going to replace Obamacare. And I'm replacing it with my own plan. And even in Massachusetts when I was governor, our plan there deals with pre-existing conditions and with young people," Romney said. "I'm not getting rid of all of healthcare reform. Of course there are a number of things that I like in healthcare reform that I'm going to put in place."

While polls show he received no "bounce" in the aftermath of the Republican National Convention, Romney still argued that he's in a "better spot" since voters were able to become more aware of his personal story and his policies.

"I think we're making real progress," Romney said. "People go to see Ann and hear our story. And the result of that is I'm better known, for better or for worse. And that allows me to continue to hammer away on what I do to get America on the right track. And I have really two months to be able to convince people I can do a better job than the incumbent. I think I can do that. So I'm in a better spot than I was before the convention."

Romney seized on Friday's disappointing jobs report as proof that he has a good chance of defeating Obama in November.

"I think it's tough to beat an incumbent if the incumbent's record is good," Romney said. "I think this incumbent has a very challenged record?This president has not been able to deliver on his promises. People are dissatisfied with where he's taken the country. And that gives me an opportunity which might not have been available had he done what he said he would do."

Asked if he would be willing? to "endanger yourself politically to the point that you were a one-term president," Romney said he "could not care less about his political prospects."

"I want to become president of the United States to get this country on the right track again. America is at a critical crossroads," the GOP candidate said. "We've got to put Americans back to work. And politics, whether I'm highly favored, not highly favored, just doesn't enter into the equation."

A survey conducted by Emily Matthew over on the Pricecharting blog, found that 80% of 874 respondents polled believe sexism is rampant in the gaming community, and 35% have been on the receiving end of sexual harassment while playing online.

Matthew?s study found that women were harassed four times more than men, and 63% of women reported being called a ?c*nt, bitch, slut, and whore.? Others reported they were threatened with sexual assault, or asked for sexual favors, and stereotypical comments regarding female gender roles were prevalent as well. We?re assuming these comments include, but were not limited to: ?go make me a sandwich? or ?get back in the kitchen and make me some pie.?

Many insults hurled at the women, according to the survey participants, also included derogatory comments regarding their weight or general physical appearance.

?Women were also much more likely to quit playing a game because of sex-based harassment than were men,? says the report. ?35.8% of women reported having quit playing temporarily because of sexism, and 9.6% reported that they quit playing a certain game permanently because of harassment.

?The numbers for men in the same areas were 11.7% and 2.6% respectively ? about a third of the percentage for women in each case.?

Sexual harassment of men might sound surprising, but look at these numbers: ?15.7% of men also reported that they had experienced sex-based taunting, harassment, or threats while playing video games.?

Granted, this figure can be construed as a ?minority? but it is still sexism.

?The comments directed at these gamers, however, are different from those directed at women in some very telling ways,? the report continues. ?Most of the men who provided additional information on their ?yes? response to this question experienced comments that revolved around them not fitting a masculine gender role. These men were often called ?fags? and compared to or told that they were women and labeled with stereotypically feminine words.

?Those who identified as intersexed, identified with a sex that was not listed, or did not identify with any sex, the sexual harassment that was experienced largely related to not fitting into any norm. Those participants in these demographics had almost all experienced intentional misgendering from other players.?

The number of respondents who experienced harrassment outside of the game came in at 9.8% of respondents that continued outside the game, and 68% of women reportedly have kept their sexual identity secret on occasion in order to avoid sexual harassment. 13% of men said they have lied about their gender for the same reason.

Some women admitted to having used their gender as a way to acquire items, attention or favors. However, the number wasn?t that large, as only 10% of women admitted it.

?The comments and data from these two questions point to an interesting conclusion: some male gamers use the stereotype of a female ?attention whore? to their benefit by pretending to be female in order to garner special benefits,? said the report. ?Many of these men even kept images of women that they found on the internet in order to supply those gamers who helped them with nude photos and proof that they were female.

?In essence, an individual using femaleness to attain special favors and gifts from others while playing video games is more likely to be a self-identified male posing as a woman than to actually be female.?

Sounds like it?s time for everyone to grow up. If we are expecting the content of our games to show maturity, stop sexualizing/victimizing women, and stop adhering to outdated stereotypes and testosterone infused storylines, we need to start with ourselves first and with society in general. Otherwise, developers won?t take our foot stamping seriously.

Gamers scream, cry, and pull their hair like toddlers fitting over the need for mature, ?grown up? themes in video games, yet they ignore online bullying. Gamer?s rejoiced when BioWare added sexual relationship options to Mass Effect, and stood up for it when Fox News distastefully, and purposely likened it to pornography. Gamers scream the loudest when atrocities committed by mass murderers are blamed on video games by politicians and the media. They wail and sign petitions when certain genres are being targeted for ban by government officials pandering to constituents with a ?save the children? mentality.

Yet we don?t chastise or alienate those who bully other gamers online. Again, if we can?t act like adults instead of misogynistic, homophobic neanderthals, how can we expect the games industry to get with the times if we can?t?
